Abstract:

Objective    To investigate the expression of p15 protein，mRNA and its correlation with the clinical pathological characteristics in oral squamous cell carcinoma（OSCC）. Methods    The expression of p15 protein，mRNA was examined in tissues from 10 normal oral mucosa and 108 cases of oral squamous cell carcinoma by SP immunohistochemistry and real-time PCR. Results    The positive expression rate of p15 protein and p15 mRNA in OSCC was significantly lower than that in normal tissues（P < 0.05）. The average level of p15 protein in stage Ⅲ-Ⅳwas significantly lower than that in stageⅠ-Ⅱ（P < 0.01）. Patients with node metastasis had significantly lower expression rate of p15 protein than those without node metastases（P < 0.05）. Conclusion    The expression of p15 gene is reduced significantly in OSCC tissues and is closely correlated with TNM stage and node metastasis. p15 may play an important role in the progression of OSCC.

摘要：

目的    探讨p15蛋白、mRNA的表达与口腔鳞癌发生发展的关系。方法    选取2000—2011年间在上海交通大学医学院附属同仁医院口腔科手术切除的口腔鳞癌标本108例，采用免疫组化法检测10例正常口腔黏膜组织及口腔鳞癌组织中p15蛋白的表达， RT-PCR测定p15 mRNA表达。结果    口腔鳞癌组织中，p15的阳性表达率（37.96%）较正常口腔黏膜组织明显降低（P < 0.05），在Ⅰ~Ⅱ和Ⅲ~Ⅳ期中口腔鳞癌组织中p15阳性率分别为59.46%和26.76%（P < 0.05），在无颈淋巴结转移组p15阳性率（42.86%）显著高于伴有颈淋巴结转移组（20.83%）（P < 0.05），p15的mRNA表达水平在肿瘤组中较正常组织明显降低（P < 0.05）。结论    p15在口腔鳞癌中呈低表达，其与口腔鳞癌病期进展和颈部淋巴结转移有一定关系，在口腔鳞癌发生发展中可能起一定作用。p15可作为反映口腔鳞癌生物学行为和估计患者预后的客观指标。
