牙龈炎症对环孢菌素A诱发大鼠牙龈增生影响研究

摘要:

目的    研究牙龈炎症对环孢菌素A(CsA)致牙龈增生的影响及转化生长因子-β1(TGF-β1)在其中的作用。方法    2006年5—8月于南京军区福州总医院比较医学科,将34只雄性SD大鼠随机分为4组,其中对照组7只,胃饲橄榄油9周后处死;结扎组7只,右侧下颌第一磨牙牙颈部栓结正畸用细钢丝,胃饲橄榄油9周后处死;单纯用药组10只,胃饲CsA橄榄油溶液(每天30 mg/kg)9周后处死;结扎+用药组10只,右侧下颌第一磨牙牙颈部栓结正畸用细钢丝,胃饲CsA橄榄油溶液 9周处死。取下颌第一磨牙做病理切片,苏木素-伊红(HE)染色观察牙周组织变化;酶联免疫吸附试验(ELISA)检测大鼠血清中TGF-β1表达变化。 结果    胃饲橄榄油或CSA橄榄油溶液9周,结扎+用药组大鼠牙龈增生最明显,其舌侧牙龈结缔组织面积为(294705.30±2490.13)μm2,明显高于单纯用药组[(48710.54±3695.88)μm2]、结扎组[(21440.15±2735.85)μm2]和对照组[(17911.53±1672.70)μm2](P < 0.05);结扎+用药组和单纯用药组大鼠血清中TGF-β1质量浓度分别为(276.60±99.79)ng/mL和(320.11±68.27) ng/mL,均明显高于结扎组[(77.80±54.57)ng/mL]和对照组[(67.63±41.55)ng/mL](P < 0.05)。结论    在炎症存在下,CsA诱发的牙龈增生会更明显;TGF-β1在CsA诱发的牙龈增生中起一定的作用。

Abstract:

Objective    To investigate the histomorphometrical alterations and serum levels of transforming growth factor β1 (TGF-β1) in CsA-induced GO with or without gingival inflammation. Methods    A total of 34 male Sprague-Dawley rats were divided into 4 groups: Group I(control);Group II(ligation);Group III(CsA);Group IV(ligation and CsA). After 9 weeks the rats were sacrificed. The morphological examination was made and the histological change with hematoxylin and eosin(HE) staining was observed. TGF-β1 levels in serum were detected by enzyme-linked immunosorbent assay(ELISA). Results    After 9-week CsA treatment, remarkable weight loss and gingival overgrowth were found in the test rats, while those rats in which gingivitis preexisted showed more severely. TGF-β1 level in CsA-exposed groups was higher than that in untreated ones,but ligation did not affect TGF-β1 level. Conclusion    The results of this study suggest that CsA-induced GO will be more apparent in the presence of local inflammation. TGF-β1 may be a key factor for the development of GO.
