Preliminary study of osteogenic effects of HAG/PDLSC complex in bone reconstruction of alveolar cleft in rabbits

Abstract: Abstract:Objective To evaluate the osteogenic function of hyaluronangel combined with periodontal ligament stem cells on reconstruction of alveolar cleft in rabbit. Methods Alveolar cleft was surgically made in 24 rabbits,They were treated with HAG/PDLSCs in the experimental groups. Single periodontal ligament stem cells and single HAG carrier application served as control groups. Those with nothing added to the bone defects were regarded as blank controls. Histopathologic method,immunohistochemical staining, and histomorphometry were used to evaluate the osteogenic effects in the above groups. Results There was no significant difference between the control group and blank control group. However,significant difference was verified in the HAG/PDLSCs group. Conclusion It is verified that HAG/PDLSCs can promote osteogenesis in rabbit alveolar cleft reconstruction. PDLSC has the positive functions in alveolar osteogenesis.

摘要: 目的 研究牙周膜干细胞(PDLSCs)移植促进兔牙槽突裂区骨再生。方法 本研究于2010年9月至2012年9月在广州医学院中心实验室完成。选择健康成年新西兰大白兔24 只,建立单侧完全性牙槽突裂动物模型后,随机分为4组:复合体组[透明质酸钠凝胶(HAG)/PDLSCs复合体移植]、PDLSCs组(单纯植入PDLSCs)、HAG组(单纯植入HAG)和空白对照组。通过HE染色、免疫组化染色和骨计量学研究分析各组骨修复效果。结果 术后3个月时,组织学观察发现,复合体组:新形成骨内有大量骨小梁密集,较多的类成骨细胞沿着骨小梁排列,可见改建中或趋于成熟的骨单位;PDLSCs组:新生骨组织内骨小梁较密集,可见改建中的骨单位及类成骨细胞;HAG组:骨小梁稀疏,可见改建中的骨单位及沿着骨小梁排列扁平柱状成骨细胞;空白对照组:无明显新生骨组织。复合体组与其他各组相比,各指标差异均有统计学意义(P < 0.05);而PDLSCs组、HAG组及空白对照组在骨组织形成方面差异均无统计学意义(P > 0.05)。结论 HAG/PDLSCs复合体可明显促进牙槽骨再生修复重建,该复合体具有良好的临床应用前景。
