Abstract:
Objective To study the adhesion of Candida albicans to human oral epithelial keratinocytes and the affecting factors. Methods The first step was the cultivation of candida albicans standards strains and human oral epithelial keratinocytes,then different phenotypes of Candida albicans were co-cultured with human oral epithelial keratinocytes at different concentration, temperature and pH, and last,gram's staining was made;observe the adhesion test result under oil microscope and calculate adhesion index and adhesion rate. Results The adhesion rate of hyphae Candida albicans was much higher than spore(P < 0.05);as the concentration of Candida albicans increased,adhesion rate increased as well;adhesion rate was highest when pH was 6.0 to 8.0. Adhesion rate of 37℃ was higher than that of 25℃(P < 0.05). Conclution Increased adhesion rate of Candida albicans is associated with the formation of hyphae;the conditions which help the formation and growth of hyphae would increase the adhesion rate.

摘要:
目的 研究白色念珠菌对人口腔黏膜上皮角质细胞(human oral keratinocytes,HOK)的黏附作用及影响因素。方法 本研究于2012年8月至2014年3月在中国医科大学附属口腔医院中心实验室完成。体外培养白色念珠菌和HOK,在不同浓度、温度和pH值条件下,用白色念珠菌浊悬液感染HOK,并进行革兰染色,油镜下观察,计算黏附指数、黏附率。 结果 在同一浓度下,菌丝相的白色念珠菌黏附率明显高于孢子相的;随着白色念珠菌浓度的升高,黏附率也有所增加。另外,白色念珠菌浓度为107 CFU/mL与HOK的细胞浓度为105个/mL时黏附基本达到饱和,可作为后续试验的最佳浓度。随着温度升高,白色念珠菌的黏附率也随之增加。黏附的最适宜pH值范围是6.0~8.0。结论 白色念珠菌黏附率的增加与菌丝的形成有关,有利于菌丝形成和生长的条件均可提高黏附率。
