苯妥英钠对内毒素作用下人牙周膜干细胞在牙骨质片上附着影响研究

Abstract:

Abstract:Objective To evaluate the effects of PHT on the attachment of human periodontal ligament stem cells on cementum slices under existing lipopolysaccharide(LPS). Methods Attachment of hPDLSCs on the cementum slices and alteration of ultrastructural organization were observed by scanning electron microscope and transmission electron microscope under the action of 20μg/mLPHT + 100 ng/mL LPS and 100 ng/mL LPS,respectively. Results hPDLSCs in the group of 20μg/mLPHT + 100 ng/mL LPS had more plentiful cell organs and rough endoplasmic reticulum distended. Golgi complex flourishing became more prosperous,and the amount of chondriosome and ribosome increased when cultured on the fourth day;when cultured on the seventh day and twenty-first day,these hPDLSCs grew better,and had more and massive pseudopodia and adhered to cementum slices more closely;in addition,calcium deposition around hPDLSCs and collagen synthesis became more active. Conclusion The attachment and development on the cementum slices of hPDLSCs can be promoted by PHT.

摘要:

目的 观察苯妥英钠(PHT)对内毒素作用下人牙周膜干细胞(hPDLSCs)在牙骨质片上附着的影响。 方法 本研究于2012年6月至2013年2月在滨州医学院中心实验室和西安交通大学医学院中心实验室完成。利用扫描电镜和透射电镜观察hPDLSCs分别在20 μg/mL PHT+ 100 ng/mL脂多糖(LPS)与100 ng/mL LPS作用下在牙骨质片上的附着情况以及细胞超微结构的变化。 结果 与细胞培养液中加入100 ng/mL LPS比较,培养液中加入20μg/mL PHT + 100 ng/mL LPS培养72 h后hPDLSCs胞浆内细胞器更丰富,粗面内质网扩张,高尔基复合体发达,有大量的线粒体和核糖体;培养第7 d和第21 d,细胞生长更密集,伪足较多且粗大,与牙骨质片的附着更紧密,细胞周围钙盐沉积以及胶原蛋白合成更加活跃。结论 PHT可促进hPDLSCs在牙骨片上的附着和生长。
