葡萄糖酸氯己定对牙龈卟啉单胞菌结合华卟啉钠的影响研究

摘要: 目的    探讨葡萄糖酸氯己定(chlorhexidine gluconate,CHX)对牙龈卟啉单胞菌结合华卟啉钠(sinoporphyrin sodium,DVDMS)的影响。方法    根据牙龈卟啉单胞菌菌液中加入DVDMS的质量浓度分为4个实验组(10、20、40、80 μg/mL DVDMS组)与1个对照组(不加DVDMS),每组再根据加入CHX的质量分数(0、0.05%、0.10%)分为3个亚组,分别记为PBS、0.05% CHX、0.10% CHX亚组。使用光谱仪检测在入射光为405 nm时各组的光谱数据,导入Origin 7.0软件中进行处理,绘制荧光光谱曲线图,并计算各实验组累计相对荧光强度。结果 同一质量分数CHX的各实验组随着DVDMS质量浓度的增加,累计相对荧光强度均有上升趋势。在同一质量浓度DVDMS组中,各不同质量分数CHX亚组累计相对荧光强度总的比较,差异均有统计学意义(10、20、40、80 μg/mL DVDMS组的F值分别为531.94、1259.04、3585.47、8611.56,均P < 0.05);其中,0.05% CHX亚组的累计相对荧光强度大于0.10% CHX亚组,0.10% CHX亚组的累计相对荧光强度大于PBS亚组,差异均有统计学意义(均P < 0.05)。结论  质量分数为0.05%和0.10%的CHX均可增加牙龈卟啉单胞菌与DVDMS的结合,0.05% CHX的作用效果更佳。

Abstract: Objective    To investigate the effect of chlorhexidine gluconate(CHX)on sinoporphyrin sodium(DVDMS) bound by Porphyromonas gingivalis. Methods    Four experimental groups(10,20,40,80 μg/mL DVDMS group)and one control group (without DVDMS) were divided according to the concentration of DVDMS added into the Porphyromonas gingivalis bacterial solution. Then each group was divided into three subgroups according to the concentration of CHX added(0,0.05% and 0.10%),which were PBS subgroup,0.05% CHX subgroup and 0.01% CHX subgroup. Spectrometers were used to detect the spectral data of the samples when the incident light was 405 nm. The fluorescence spectrum curve was drawn by Origin 7.0 software,and the cumulative relative fluorescence intensity values of DVDMS groups were calculated. Results    For each experimental subgroup with the same concentration of CHX,the cumulative relative fluorescence intensity increased with the increase of DVDMS concentration. In DVDMS group with the same concentration,the total comparison of CHX subgroups with different concentration showed statistically significant differences(F values of 10,20,40 and 80 μg/mL DVDMS group were 531.94,1259.04,3585.47 and 8611.56,respectively,P < 0.05). Furthermore,the cumulative relative fluorescence intensity of 0.05% CHX subgroup was greater than that of 0.10% CHX subgroup,and the cumulative relative fluorescence intensity of 0.10% CHX subgroup was greater than that of PBS subgroup,the differences being statistically significant(P < 0.05). Conclusion    0.05% CHX and 0.10% CHX can increase the binding between Porphyromonas gingivalis and DVDMS,and the effect of 0.05% CHX is better.
