钙磷涂层处理对镁合金植入体周围软组织Ⅰ型胶原mRNA表达影响动物实验研究

摘要: 目的    探讨钙磷涂层对兔下颌骨镁合金植入体周围软组织中Ⅰ型胶原mRNA表达的影响。方法    本研究于2008年12月至2009年8月在中国医科大学实验动物中心和中心实验室进行。选择雄性成年日本大耳白兔27只,随机分为T组、A组和B组,每组9只。将钙磷涂层镁合金螺钉(T组)和无处理镁合金螺钉(A组)植入兔的下颌骨内,以钛合金(B组)作为对照组,每组观察时间分别为1、2、3个月。采用反转录聚合酶链反应(RT-PCR)方法检测植入体周围软组织中Ⅰ型胶原的mRNA表达情况。结果    T组Ⅰ型胶原的mRNA表达水平高于A组,随观察时间的增加,两组Ⅰ型胶原的mRNA表达量均逐渐增高。观察2和3个月时的组间差异有统计学意义(P < 0.05),观察1个月时的组间差异无统计学意义(P > 0.05);T组观察2和3个月的Ⅰ型胶原mRNA表达水平与1个月相比,均有统计学意义(P < 0.05)。结论    钙磷涂层能够提高镁合金植入体周围软组织Ⅰ型胶原的mRNA表达水平,促进早期的成骨过程,显示出良好的生物活性。

Abstract: Objective    To investigate effect of calcium phosphate on typeⅠcollagen expression of soft tissue around magnesium alloy in rabbit mandible. Methods    This study was finished in the Experimental Animal Center and Central Laboratory of China Medical University from December 2008 to August 2009. Twenty-seven healthy adult male Japanese white rabbits were randomly divided into calcium phosphate coating group,untreated group and titanium alloy group. we implanted calcium phosphate coating and untreated AZ31B magnesium alloy screws into the rabbits’ mandible,titanium alloy as the control. The animals were sacrificed after 1 month,2 months and 3 months respectively;typeⅠcollagen mRNA expressions were measured by RT-PCR. Results    The typeⅠcollagen mRNA expressions of coating group was higher than untreated group. With time passing,the typeⅠcollagen mRNA expression showed a gradual increase for each group,there being significant differences between groups at 2nd and 3rd month(P < 0.05). Significant differences exsited between 1st month and 2nd,3rd month of coating group(P < 0.05). Conclusion    Calcium phosphate coating improves the mRNA expression of typeⅠcollagen of soft tissue around magnesium alloys and is beneficial to the early osteogenesis,manifesting good biological activity.
