不同时段牙周膜牵张成骨快速移动牙齿对牙髓中TNF-α表达影响研究

摘要:

目的    研究牙周模牵张成骨速移动牙齿时牙髓组织中肿瘤坏死因子α(TNF-α)含量变化情况。方法    2009年12月至2010年2月选用山东大学实验动物中心提供健康杂种犬18只,随机分为9组,每组2只。以单根的第一前磨牙作为移动牙,通过牙周膜牵张成骨快速移动牙齿,分别在加力2(A1组)、4(A2组)周及各自加力后保持1(B1、B2组)、2(C1、C2组)、4(D1、D2、E组,其中E组为对照组,不加力)周时,取出牙髓组织标本,利用酶联免疫吸附试验检测牙髓组织中TNF-α含量。结果    TNF-α含量在牵张加力2、4周后明显升高,停止牵张加力后第1、2周TNF-α含量明显降低,第3、4周TNF-α含量稳定在一定水平上,其含量低于对照组。结论    牙槽骨牵张2周及4周快速移动牙齿时,牙髓内TNF-α含量均可在牵张后2周内基本恢复正常,而牵张后7~14 d则为其转变的关键时期。

Abstract:

Objective    To observe the changes of TNF-α in the dental pulp by using distraction osteogenesis on orthodontic rapid tooth movement after two and four weeks. Methods    Eighteen native healthy-hybrid dogs were randomly divided into 9 groups from December 2009 to February 2010. Two dogs were in each group. The first premolar of the experimental animals was moved by using the distraction device and then was extracted when the device had been activated for 2(A1 group)and 4(A2 group) weeks and fixed for 1 week(B1,B2 group),2 weeks(C1,C2 group)and 4(D1,D2,E group,E group was control group) weeks respectively. Then evaluate the changes of TNF-α in the dental pulp by using distraction osteogenesis on rapid tooth movement during different periods;the quantity of TNF-α in different stages were detected by ELISA methods. Results    The quantity of TNF-α increased greatly right after two and four weeks’application of distraction force, while it significantly decreased during the first week and the second week after stopping distraction osteogenesis,and the quantity of TNF-α changed little from day 14 to 28 after stopping distraction osteogenesis,but the TNF-α level from day 14 to 28 was lower than that of the control group. Conclusion    The quantity of TNF-α can return to normal in two weeks after two and four weeks’distraction force applied to the orthodontic tooth;the period from day 7 to 14 is the key period for TNF-α inflammation 's transformation.
