Rho激酶抑制剂Y-27632对涎腺腺样囊性癌细胞生长增殖影响的研究

摘要：

目的    观察Rho激酶抑制剂Y-27632对涎腺腺样囊性癌细胞肺高转移细胞株（ACC-M）生长增殖的影响，探讨Y-27632对涎腺腺样囊性癌可能疗效，以期为治疗涎腺腺样囊性癌提供新思路。方法    2007年10月至2008年2月在浙江大学医学实验室，形态学观察Rho激酶抑制剂Y-27632作用下ACC-M细胞的生长变化，并用四甲基偶氮唑蓝（MTT）比色法检测不同药物浓度下ACC-M的增殖情况。结果    抑制剂Y-27632作用下ACC-M细胞变成梭形，细胞长轴增加，细胞间连接消失，细胞膜不规则，皱缩不整齐，边界有丝状突起，有倾向凋亡的趋势；MTT比色法检测结果发现，不同浓度Y-27632作用下检测到的ACC-M细胞光密度值均比对照组小，且当药物浓度为10 μmol／L时抑制作用有统计学意义（P < 0.01）。结论    Rho激酶抑制剂Y-27632在体外实验中对涎腺腺样囊性癌细胞的生长增殖起一定的抑制作用。

Abstract:

Objective    To investigate the effects of Rho-kinase inhibitor Y-27632 on the development and proliferation of adenoid cystic carcinoma cells，so as to provide new ideas for the treatment of adenoid cystic carcinoma. Methods         Morphology change：observe the ACC-M cells after 10 μmol／L Y-27632 added in medium；Methabenzthiazuron tetrazolium（MTT）：ACC- M exposed to various concentration of Y-27632（0、2.5、5、10、20 μmol／L）24 hours，then terminate and detect the OD value. Results    The morphology changed a lot in cells of ACC-M when exposed to 10 μmol／L Y-27632. With MTT we found cell proliferation of ACC- M was inhibited significantly at 10 μmol／L （P < 0.01）. Conclusion    The inhibitor Y-27632 can do some inhibition to the proliferation of ACC in vitro.
