NS398对SACC-83细胞抑制作用实验研究

摘要:

目的    研究环氧合酶抑制剂NS398对唾液腺腺样囊性癌(SACC)-83细胞的抑制作用。方法    2008年6—12月于中国医科大学中心实验室,用含0、50、100、150、200 μmol/L NS398的培养液体外培养SACC-83细胞24、48、72、96 h后,应用噻唑蓝(MTT)快速比色法,检测NS398对SACC-83细胞的抑制作用;用上述实验确定的最大抑制浓度培养液培养细胞,采用倒置相差显微镜和透射电镜观察细胞形态。结果    随着作用时间延长和药物浓度增加,NS398对SACC-83细胞的抑制率逐渐增加。NS398在浓度150 μmol/L作用72h后,抑制作用最明显。倒置相差显微镜观察,随着培养时间的增加,试验组(150 μmol/L NS398)贴壁细胞数量明显减少,胞浆开始脱水产生空泡,并与胞膜融合,出现胞膜空泡化,胞核深染,胞质浓缩;透射电镜观察,试验组(150 μmol/L NS398)细胞出现凋亡现象。结论    NS398可抑制SACC-83细胞的增殖,并具有时间和浓度依赖性,NS398可能在唾液腺腺样囊性癌治疗中发挥重要作用。

Abstract:

Objective    To investigate the inhibitive effect of COX-2 inhibitor NS398 on cancer cell growth of salivary adenoid cystic carcinoma SACC-83 cell. Methods    The SACC-83 cell culture median contained 0,50,100,150,200μmol/L NS398,and individually cultured for 24,48,72,96 hours. MTT assay was used to study the inhibitive effect of NS398 on the growth of SACC-83 cells, and then inverted phase contrast microscope and transmission electron microscope were used to observe the cell morphological changes. Results    The inhibitive effect increased with the time prolonging and the concentration increasing. The effect was most obvious with 150μmol/L NS398 cultured for 72 hours(concentration: P<0.05,time: P<0.05). With the culture time prolonging, observed under inverted phase contrast microscope, the test group(150 μmol/L NS398) showed significant reduction in attached cells, cytoplasmic vacuolization, deeply stained nuclei and cytoplasmic concentration. Observed under the transmission electron microscope, the test group (150 μmol/L NS398) showed apoptosis. Conclusion    NS398 can inhibit the growth of SACC-83 cells. The inhibitive effect of NS398 on SACC-83 cells is dose-dependent and time-dependent.NS398 may play an important role in the treatment of salivary adenoid cystic carcinoma.
