面神经损伤诱导面神经元死亡及其调控基因表达的实验研究

摘要:

目的 研究大鼠面神经总干离断伤后神经元的形态学改变及对TUNEL、Fas和Caspase-3、Caspase-8表达的影响。方法 2009年1 — 8月于中国医科大学实验技术中心手术制作大鼠右侧面神经总干切断伤模型,左侧为正常对照。伤后各时间点,通过HE染色观测面神经核神经元的形态学变化;TUNEL检测细胞凋亡;免疫组化检测Fas和Caspase-3、Caspase-8表达的变化。结果 伤后3 d开始面神经元存活率逐渐下降,28 d时存活率达最低;伤后第7d开始可见TUNEL阳性表达,14 d时数量达高峰;伤后1 d时,Fas表达增高,于7 d时达高峰。伤后3 d时,Caspase-3、Caspase-8表达增高, 14 d时达高峰。结论 面神经总干离断伤可引起面神经元凋亡;Fas和Caspase-3、Caspase-8参与面神经总干离断伤后诱导其神经元凋亡过程的调控。

Abstract:

Objective To investigate the morphology of facial neurons and the expression of TUNEL、Fas and Caspase-3、-8 following facial nerve transaction in rats. Methods The right facial nerve was transaction at its stylomastoid foramen,and the left side as normal control . Each time point after injury, the morphology of facial neurons were observed under light microscope; the apoptotic cells of injured FMNs were detected by TUNEL staining;the expression of Fas,Caspase-3 and Caspase-8 were detected by immunohistochemistry method. Results The survival rates of FMNs decreased gradually from the 3rd day and lowest on the 28th day after injury. TUNEL-positive cell was noted on the 7th day after injury and its number peaked on the 14th day. The expression of Fas increased at the 1st day after injury and peaked at the 7th day. The expression of Caspase-3 and Caspase-8 increased at the 3rd day after injury and peaked at the 14th day. Conclusion Facial nerve transaction can cause facial neurons apoptosis .Fas,Caspase-3 and Caspase-8 might play an important regulating role in the process of facial neurons apoptosis following facial nerve transaction.
