口腔临床常用烤瓷合金对小鼠肺成纤维细胞体外毒性研究

Cytotoxicity of dental ceramic alloys on L929 mouse fibroblasts:an in vitro study

摘要: 目的    比较镍铬、钴铬、纯钛、钯基、金铂等5种常用烤瓷合金的细胞毒性。方法    本研究于2011 年7—10 月在福建医科大学附属口腔医院中心实验室进行。按照ISO10993标准,采用CCK-8法测定第1、3、5、7天时烤瓷合金浸提液培养的小鼠肺成纤维细胞(L929细胞)的OD值,同时观察L929细胞形态,并且计算细胞相对增殖率(RGR),评价5种烤瓷合金对L929的细胞毒性。结果    第7天时,除金铂合金外,其他4组烤瓷合金的OD值与阴性对照组的差别均有统计学意义(P < 0.05);L929细胞的RGR由高到低分别为金铂合金(98.4%)、钯基合金(95.2%)、纯钛合金(83.0%)、镍铬合金(74.0%)、钴铬合金(72.6%)。细胞形态学观察发现:金铂合金组、钯基合金组以及纯钛合金组的细胞形态与阴性对照组相比无明显差别;而钴铬合金组和镍铬合金组的细胞排列稀疏,少量细胞皱缩变圆。结论    金铂合金、钯基合金以及纯钛合金的细胞毒性为1级,具有较好的生物相容性;钴铬合金、镍铬合金的细胞毒性为2级,结合细胞形态观察认为生物相容性仍然合格。

Abstract: Objective    To evaluate the cytotoxicity of Au-Pt,Ni-Cr,CP Ti,Co-Cr and Pd-based ceramic alloys on L929 mouse fibroblasts in vitro. Methods    According to the ISO 10993,L929 mouse fibroblasts were cultured with elute of the ceramic alloys. Optical density(OD)was analyzed with cell counting kit-8(CCK-8)assay on day 1,3,5 and 7. The morphology of L929 was evaluated. Cellular relative growth rate(RGR)was calculated to evaluate the cytotoxicity. Results    On day 7,OD values in all test groups,except for Au-Pt group,were significantly lower than the negative controls(P < 0.05). The RGR values were ranked as follows:Au-Pt(98.4%),Pd-based(95.2%),CP Ti(83.0%),Ni-Cr(74.0%)and Co-Cr(72.6%). The cells of Au-Pt,CP Ti and Pd-based group had no significant morphological difference compared to the negative controls. The cells of Ni-Cr group and Co-Cr group were sparsely arranged,which were a bit shrunk compared to the negative controls. Conclusion    Cytotoxicity level grade 1 is found in Au-Pt,CP Ti and Pd-based ceramic alloys,which is considered to have good biocompatibility. Cytotoxicity level grade 2 is found in Co-Cr and Ni-Cr ceramic alloys. Combined with the results of cell morphology observation,Co-Cr and Ni-Cr ceramic alloys are considered qualified for clinical usage as for biocompatibility.
