Abstract: AbstractObjectiveThe main purpose of the study was to research into the correlation of cord blood leptin levels and neonates immunologic function as well as the possible immunomodulate mechanism. MethodsThirty preterm infants and 60 term infants of our hospital were selected from March to October in 2003,respectively.They were checked up the contents of leptin,GMCSF,IgG,IgA,IgM,C3 and C4 in cord blood serum,and the expression of IL4,IFNγ,bcl2 protein,T lymphocyte subpopulation and lymphocyte apoptosis percentage in cord blood by radioimmunoassay(RIA)、immunotransmission turbidity method and flowcytometry. ResultsThe umbilical vein blood leptin contents were positively correlated with the IgG (P<001)、C3 (P<001)、C4 (P<005) levels、the CD4+T lymphocyte percentage (P<005)、CD8+T lymphocyte percentage (P<001)、IFNγ levels (P<005)、GMCSF levels and the expression of bcl2 protein;there had negative correlations between the cord blood leptin levels and IL4 levels、lymphocyte apoptosis percentage;there had no significant correlation between leptin levels and IgA、IgM levels (P>005). ConclusionThe cord blood leptin levels are strongly correlated with neonates’ immune functions.Leptin could enhance the cellular immune function.But leptin has minor influence on humoral immunity.Leptin could raise the expression of bcl2 albumin and the contents of GMCSF,which is believed to be the possible important mechanisms of the leptin's immunomodulation function.

摘要: 目的探讨脐血瘦素(leptin)质量浓度与新生儿免疫功能的关系及其可能的免疫调控机制。 方法2003年3~10月河北医科大学第四医院儿科对正常分娩的早产儿30例、足月儿60例,分别采用放射免疫分析(RIA)、免疫透射比浊法及流式细胞术(FCM)检测脐血leptin、粒细胞巨噬细胞集落刺激因子(GMCSF)、IgG、IgA、IgM、补体(C3、C4)、白细胞介素4(IL4)、γ干扰素(IFNγ)质量浓度及bcl2蛋白表达量、T淋巴细胞亚群、淋巴细胞凋亡率。 结果脐血leptin质量浓度与IgG(r=0630,P<001)、C3(r=0340,P<001)、C4(r=0253,P<005)、CD+4T淋巴细胞百分率(r=0295,P<005)、CD+8T淋巴细胞百分率(r=0377,P<001)及IFNγ质量浓度(r=0323,P<005)、GMCSF质量浓度(r=0236,P<005)、bcl2蛋白表达量(r=0318,P<005)均呈正相关,与IL4质量浓度(r=-0277,P<005)、淋巴细胞凋亡率(r=-0532,P<001)呈负相关,而与IgA、IgM无显著相关(均P>005)。 结论脐血leptin质量浓度与新生儿免疫功能密切相关,leptin具有增强新生儿细胞免疫功能的作用,而对体液免疫功能影响较小。leptin可上调bcl2蛋白表达和GMCSF质量浓度,抑制淋巴细胞的凋亡,此作用可能是leptin免疫调节作用的重要机制。
