微弧氧化处理Ti2448的生物相容性研究

Abstract:

Objective To evaluate the biocompatibility of Ti-24 Nb-4 Zr-7.9 Sn(Ti2448)by Micro-arc oxidation. Methods Ti2448 material was treated by Micro-arc oxidation (MAO-Ti2448). The proliferation and toxicity of mouse fibroblasts(L929)cultured in each group were compared by MTT assay. The guinea pig skin reaction of each group was observed by the sensitization test. Mouse embryo osteoblast precursor cells(MC3T3-E1)was co-cultured with materials and the alkaline phosphatase(ALP)activity of MC3T3-E1 was measured. Results The cytotoxicity grade of group MAO-Ti2448 and group Ti2448 was 0,which showed that they had no cytotoxicity. Optical density of L929 cells in group MAO-Ti2448 leaching liquor was increased along with the time,and the OD in group MAO-Ti2448 was significantly more than in group Ti2448(P < 0.05). The skin sensibility grade of group MAO-Ti2448 and group Ti2448 was 0,which showed that group MAO-Ti2448 caused no skin sensibility. ALP activity of group MAO-Ti2448 and group Ti2448 was increased along with the time. ALP activity of MC3T3 cells in group MAO-Ti2448 was significantly more than in group Ti2448,and the difference was statistically significant(P < 0.05). Conclusion MAO-Ti2448 has no cytotoxicity and it can promote the proliferation and early osteogenic differentiation. MAO-Ti2448 has good biocompatibility.

摘要:

目的 评价微弧氧化处理低弹性模量钛铌锆锡合金(Ti-24 Nb-4 Zr-7.9 Sn,Ti2448)的生物相容性。方法  通过微弧氧化处理Ti2448试件(MAO-Ti2448)。采用MTT法比较MAO-Ti2448组和Ti2448组浸提液培养小鼠成纤维细胞(L929)的增殖和毒性情况;应用致敏实验观察各组浸提液引起的豚鼠皮肤反应;各组与小鼠胚胎成骨细胞前体细胞(MC3T3-E1)共培养,测定MC3T3-E1的碱性磷酸酶(ALP)活性。结果 MAO-Ti2448组与Ti2448组的细胞毒性分级为0级,均无细胞毒性。随着细胞培养时间延长,MAO-Ti2448组和Ti2448组的吸光度(OD)值均增加,且MAO-Ti2448组的OD值明显高于Ti2448组,差异有统计学意义(P < 0.05)。MAO-Ti2448组与Ti2448组的皮肤反应均为0级,无皮肤致敏性。随着细胞培养时间延长,MAO-Ti2448组和Ti2448组MC3T3-E1的ALP活性均增高,与Ti2448组对比,MAO-Ti2448组MC3T3-E1的ALP活性较高,差异有统计学意义(P < 0.05)。结论 微弧氧化处理Ti2448无细胞毒性,可促进细胞增殖及早期成骨分化,具有良好的生物相容性。
