Abstract:
Sclerostin/SOST,mainly expressed in osteocytes,is a negative regulator of bone formation and is regulated by mechanical stimulus,which plays an important role in orthodontic tooth movement. Wnt and BMP are two important signaling pathways in bone metabolic regulation. SOST can regulate osteoblastic differentiation and bone formation by binding type ⅠorⅡreceptors and co-receptor LRP5/6 to inhibit BMP and Wnt signaling pathways. This review covers the effect of Sclerostin/SOST on orthodontic tooth movement after mechanical loading and its mechanism,and the clinical significance as well as prospect of Sclerostin/SOST in clinical application.

摘要:
骨硬化蛋白(Sclerostin/SOST)主要由骨细胞特异性表达,是骨形成的负性调节因子,受机械刺激的调控,在正畸牙移动中发挥着重要的作用。Wnt和骨形成蛋白(BMP)是正畸牙移动骨改建的两个重要信号途径,骨硬化蛋白可通过结合BMP的Ⅰ型或Ⅱ型受体和Wnt的共受体LRP5/6分别抑制BMP和Wnt信号途径来调控成骨细胞分化和骨形成。文章综述了骨硬化蛋白对机械加力后正畸牙移动的影响和作用机制及其临床意义与应用前景。
