乳腺癌组织中MCM-5和Ki-67的表达及临床意义

摘要:

目的 探讨微小染色体维持蛋白-5(minichromosome maintenance-5,MCM?5)和细胞增殖抗原标记物(Ki67)在乳腺癌组织中的表达及临床意义。方法 2007年1月至2008年4月广西医科大学第五附属医院采用免疫组织化学方法(SP法)对经手术诊断的45例乳腺癌组织中MCM?5 和Ki?67基因蛋白进行检测,分析其表达情况。 结果 MCM?5、Ki?67的表达与临床分期(P<0.05)、淋巴结转移(P<0.01)显著相关,无淋巴结转移组明显低于淋巴结转移组;Ki?67与ER、PR表达无显著相关。 结论 MCM?5、Ki?67与乳腺癌的发生发展密切相关,可作为判断肿瘤浸润转移、指导治疗和估计预后的参考指标。特别是MCM?5的表达水平可能是判断乳腺癌的术后生存的独立有效指标,而综合应用上述指标可能更有助于判断预后。

Abstract:

The Fifth Affiliated Hospital, Guangxi Medical University, Liuzhou 545001, China Corresponding author:HUANG Ping, E-mail:mojunyang2006@163.com Abstract Objective To study the expressions and clinical significance of minichromosome maintenance?5 (MCM?5) and Ki-67 in breast cancer tissue.Methods Immunohistochemistry (SP method) was applied to detect the expression of MCM-5 and Ki-67 genes in tissues from 45 cases of breast cancer after operation from January 2007 to April 2008 at the Fifth Affiliated Hospital of Guangxi Medical University.Results The expression level of MCM-5, Ki-67 was significantly correlated with clinical stage (P<0.05) and axillary lymph node metastases (P<0.01). The expression level of MCM-5 and Ki-67 in no lymph node metastasis cases was higher than those in lymph node metastasis cases significantly. The expression of Ki-67 was not correlated with ER and PR status. Conclusion The gene products of MCM?5and Ki-67 play a key role in the oncogenesis and development of breast cancer. Their expression could be useful indexes to assess the malignant level and clinical prognosis of breast cancer. The expression of MCM?5 may be a independent prognostic factor of postoperative survival for patients with breast cancer.The combined use of the above markers may be helpful in making a better estimation of prognosis.
