Abstract:
Abstract:Objective To study the expression and correlation of PTEN, MMP-7 and VEGF in oral squamous cell carcinoma. Methods The expression of PTEN, MMP-7 and VEGF in 40 cases of OSCC tissues and 40 cases of normal oral mucosa tissues were detected using immunohistochemical SP method, respectively. And the correlation among the expression of PTEN, MMP-7 and VEGF in OSCC were analyzed. Results The expression of MMP-7 and VEGF were higher in OSCC than in normal oral mucosa( P < 0.05) .The positive rate of PTEN protein in OSCC tissues was significantly lower than it in normal oral mucosa tissues(P < 0.05). In OSCC,the expression of PTEN was negatively correlated with the expression of MMP-7 and VEGF. Furthermore, the expression of MMP-7 was positively correlated with the expression of VEGF. Conclusion Down-regulation of PTEN expression and up-regulation of MMP-7, VEGF expression were closely related to the occurrence and development of OSCC. Measurement of the expression of PTEN,MMP-7 and VEGF may have a clinical significance in understanding malignant behavior of OSCC.

摘要:
目的 探讨人第10号染色体缺失性磷酸酶-张力蛋白同源物基因(PTEN)、基质金属蛋白酶-7(MMP-7)和血管内皮细胞生长因子(VEGF)在口腔鳞状细胞癌(OSCC)中的表达以及相关性。方法 采用免疫组化SP法,分别检测40例OSCC组织和40例正常口腔黏膜组织中PTEN、MMP-7和VEGF 的表达情况,并分析PTEN、MMP-7和VEGF在OSCC中表达的相关性。结果 PTEN、MMP-7和VEGF在OSCC组织中的表达率分别为35.0%(14/40)、65.0%(26/40)和72.5%(29/40),在正常口腔黏膜组织中的表达率分别为77.5%(31/40)、22.5%(9/40)和25.0%(10/40),3个指标在两种组织间的表达差异均有统计学意义(P<0.05)。OSCC组织中,PTEN 表达与MMP-7及VEGF 表达均呈负相关(r分别为-0.406和-0.394,均P<0.05);MMP-7表达与VEGF表达呈正相关(r = 0.514,P<0.05)。结论 PTEN 表达下调和MMP-7、VEGF表达上调与OSCC的发生发展密切相关;联合检测三者的表达对了解OSCC的恶性行为有重要的临床参考价值。
