涎腺Warthin瘤上皮VEGF-C表达及其与间质微淋巴管密度相关性研究

摘要：

目的    研究Warthin瘤中微淋巴管密度（MLVD）与肿瘤上皮细胞中血管内皮生长因子C（VEGF-C）的相关性。方法    收集1999—2008年内蒙古医学院附属医院和内蒙古自治区医院病理科存档的Warthin瘤病例109例，同时选取多形性腺瘤（PA）、正常涎腺、正常涎腺淋巴结各20例，采用免疫组化法检测各组织标本中VEGF-C和淋巴管内皮透明质酸受体-1（LYVE-1）的表达情况，并通过LYVE-1的表达来计数MLVD。结果    （1）Warthin瘤中MLVD明显高于PA、正常涎腺及其淋巴结（均P < 0.01）；（2）VEGF-C在Warthin瘤组织中呈强阳性表达，而在PA、正常涎腺组织及其淋巴结中呈弱阳性或阴性表达；（3）Warthin瘤中VEGF-C表达强度与MLVD呈正相关关系。结论    VEGF-C可介导Warthin瘤淋巴管生成。

Abstract:

Objective    To investigate the mechanism of lymphangiogenesis by immunohistochemical stain，and provide the envidence for histopathological occurrence in Warthin’s tumor. Methods    The subjects of this study were 109 patients suffering from Warthin’s tumor. At the same time，select pleomorphic adenoma（PA）20 cases，normal parotid gland 20 cases，and normal intraparotid lymph node 20 cases as control groups. The expression of VEGF-C was evaluated in each group by immunohistochemical staining two-step methods. The microlymphatic vessel density （MLVD） in Warthin’s tumor was counted by immuno-staining with the specific antibody LYVE-1. Results    The results of immunohistochemical staining：（1）MLVD was higher in Warthin’s tumor than in pleomorphic adenoma（PA），normal parotid gland， and normal intraparotid lymph node（P < 0.01）. （2）The expression of VEGF-C was significant in Warthin’s tumor. There was negative or weakly positive expression of VEGF-C on the modes of PA（P < 0.01）. （3） In Warthin’s tumor，VEGF-C expression positively correlated with the MLVD（r=0.874）. Conclusion    VEGF-C mainly promotes lymphangiogenesis in Warthin’s tumor.
