TGF-β1在表面处理钛合金种植体骨界面表达变化的实验研究

摘要:

目的    通过检测转化生长因子-β1(TGF-β1)在种植体骨界面骨组织中的表达,评价表面处理钛合金、钛合金及不锈钢种植体的生物相容性。方法    于2002年3月至2003年3月在中国医科大学组织学与胚胎学教研室随机将90只大鼠分为3组,每组30只,雌雄各半。分别将表面氧化处理后的六铝四钒钛(处理钛合金组)、未经特殊处理的六铝四钒钛(Ti6Al4V组)、不锈钢(316L合金组),植入大鼠的下颌骨内,于术后第1、2、3、4、6周取材,进行HE染色观察及TGF-β1免疫组化染色灰度值判读。结果    (1) HE染色光镜观察:处理钛合金组种植体成骨细胞排列紧密,新生骨较其余两组更致密。(2)TGF-β1免疫组化染色灰度值的比较:处理钛合金组高于Ti6Al4V组,Ti6Al4V组高于316合金组(均P < 0.05)。结论    表面氧化处理可增强Ti6Al4V骨界面TGF-β1的表达,提高其生物相容性。

Abstract:

Objective    To evaluate the biocompatibility of surface-prepared Ti6Al4V by examining expression of  TGF-β1 on the interface of implant and bone. Methods    Three kinds of materials including surface-oxidated/non-prepared Ti6Al4V and stainless steel were randomly implanted into mandibles of 90 rats, specimens were obtained 1, 2,3,4 and 6 weeks after implantation,respectively. HE stain and immunohistochemistry were used to examine the interface of implant and bone. Results    Osteoblasts arranged more closely together in surface-prepared Ti6Al4V group than the other groups by IE observation. In comparison of TGF-β1 immunohistochemistry expression, surface-prepared Ti6Al4V>non- prepared Ti6Al4V> stainless steel(P < 0.05). Conclusion    Surface oxidation may enhance TGF-β1 expression on the implant-bone interface and increase the biocompatibility of implant.
