Abstract:
Objective To compare the effects of Shikonin and Juglone on the proliferation and apoptosis of human tongue squamous cell carcinoma(Tca8113). Methods Inhibitory effect of Shikonin and Juglone was determined by MTT assay and recorded by cell growth curve,and apoptosis-induction effect of both medicines was tested by Hoechst 33258 staining. The data was analyzed by one-way ANOVA test and t-test using SPSS19.0 software package. Results Shikonin and Juglone inhibited the growth of Tca8113 cell lines in a time- and dose-dependent manner, ranging from 5 to 25 μmol/L(P < 0.05),and both medicines could induce apoptosis of Tca8113cell lines. The inhibition and induction effect of Shikonin on Tca8113 cell lines was better than Juglone at 15 μmol/L(P < 0.05). Conclusion The effect of Shikonin on the proliferative inhibition and apoptosis induction of Tca8113 is better than Juglone.

摘要:
目的 比较紫草素和胡桃醌对舌癌Tca8113 细胞的增殖抑制及凋亡诱导作用。方法 采用MTT法观察紫草素和胡桃醌对 Tca8113 细胞的增殖抑制作用并绘制细胞生长曲线;采用Hoechst 33258 染色法检测两种药物对 Tca8113细胞的凋亡诱导作用;采用 SPSS19.0 软件包对数据进行单因素方差分析及t检验。结果 5~25 μmol/L浓度范围内,两种药物对 Tca8113 细胞的生长抑制作用呈现出明显的剂量和时间依赖性,且两种药物均可诱导 Tca8113 细胞凋亡;15 μmol/L紫草素的增殖抑制作用及诱导凋亡作用均强于胡桃醌(P < 0.05)。结论 紫草素对 Tca8113 细胞的增殖抑制作用及凋亡诱导作用强于胡桃醌。
