葡萄糖浓度对人牙龈成纤维细胞分泌Ⅰ、Ⅲ型胶原蛋白及纤维粘连蛋白影响研究

摘要:

目的    探讨不同浓度葡萄糖对人牙龈成纤维细胞分泌Ⅰ(COL1)、Ⅲ型胶原蛋白(COL3)及纤维粘连蛋白(FN)的影响。方法    分别于5.5、7.0、8.8、10.0、15.0、20.0 mmol/L的葡萄糖溶液中培养人牙龈成纤维细胞3 ~ 7 d,酶联免疫吸附试验(ELISA)法测定其分泌COL1、COL3及FN的量。结果    人牙龈成纤维细胞培养3 d后,葡萄糖浓度高于10.0 mmol/L时,COL1的分泌量减少;高于15.0 mmol/L时,COL3的分泌量减少;FN与COL3情况相似。培养7 d后,葡萄糖浓度高于10.0 mmol/L时,COL1、COL3及FN的分泌量均减少。结论    随着葡萄糖浓度的增高,人牙龈成纤维细胞分泌COL1、COL3及FN的量均有减少趋势,且高糖条件下更为明显。

Abstract:

Objective    To investigate the effect of different concentrations of glucose on human gingival fibroblasts secreting collagenⅠ,collagen Ⅲ and fibronectin.  Methods    The human gingival fibroblasts were cultured in different concentrations of glucose solution for 3 days to 7 days respectively,the concentrations being 5.5,7.0,8.8,10.0,15.0 and 20.0 mmol/L. The amount of collagenⅠ,collagen Ⅲ and fibronectin secreted by the human gingival fibroblasts were measured through Elisa method. Results    When the human gingival fibroblasts were cultured for 3 days,the amount of COL1 decreased in the glucose solution whose concentration was higher than 10.0 mmol/L;the amount of COL3 decreased when the glucose concentration was higher than 15.0 mmol/L,and so was FN. When the human gingival fibroblasts were cultured for 7 days,the amount of COL1,COL3 and FN decreased in the glucose solution whose concentration was higher than 10.0 mmol/L. Conclusion    With glucose concentrations increasing,the amount of collagenⅠ,collagen Ⅲ and fibronectin decreases,and the decrease is more significant in the high concentration of glucose solution.
