不同存在形式的纤维蛋白原对中性多形核白细胞分泌IL-1β和IL-8影响的研究

摘要: 目的 探讨不同存在形式的纤维蛋白原(Fg)对人外周血中性多形核白细胞(PMN)分泌白细胞介素(IL)-1β和IL-8的影响及其在牙周炎发病机制中的病理作用。方法 2006年5月至7月,在四川大学华西口腔医学院牙周科对4名研究对象以梯度密度离心法分离外周血PMN并与可溶性或固着性的不同存在形式的Fg作用,酶联免疫吸附试验(ELISA)法测定其分泌IL-1β和IL-8的质量浓度。结果 与可溶性、固着性Fg作用后,PMN分泌IL-1β和IL-8的质量浓度差异均具有统计学意义(P < 0.05);与固着性Fg作用后,PMN分泌IL-1β和IL-8的质量浓度显著升高(P < 0.05)。结论 与固着性Fg作用可使PMN分泌高质量浓度的促炎细胞因子IL-1β和IL-8,在牙周炎发病机制中可能具有一定的病理作用。

Abstract: Objective To study the effect of different forms of fibrinogen on the secretion of IL-1β and -8 of human peripheral polymorphonuclear leukocytes (PMNs) and its pathological role in the pathogenesis of periodontitis. Methods Peripheral PMNs were isolated by discontinuous density gradient centrifuging technique. The freshly isolated PMNs were cultured in the present of different forms of fibrinogen which existed as soluble form or immobilized form, and the levels of IL-1β and -8 in the supernatants produced by PMNs were measured by ELISA technique. Results The differences in the levels of IL-1β and -8 released by peripheral PMNs between the two groups after PMNs interacted with immobilized and soluble fibrinogen, were significant(P < 0.05). Depending on the interaction with immobilized fibrinogen, but not soluble fibrinogen, PMNs secreted significantly higher levels of IL-1β and IL-8. Conclusion The interaction between PMNs and immobilized fibrinogen could induce PMNs to release higher levels of pro-inflammatory cytokines, IL-1β and IL-8, which may be involved in the pathogenesis of periodontitis.
