miR-15b对口腔鳞癌细胞Cal27增殖与凋亡能力影响研究

摘要:

目的    研究口腔鳞癌(OSCC)中miR-15b的表达及其对OSCC Cal27细胞增殖和凋亡能力的影响。 方法    实时定量PCR检测2013年1月至2015年1月期间中国医科大学附属口腔医院外科50例住院患者OSCC组织中miR-15b基因的表达水平。培养OSCC Cal27细胞,将miR-15b的前体转染Cal27细胞,MTT法及双标流式法检测转染前后Cal27细胞的增殖和凋亡能力。 结果    与癌旁组织相比,miR-15b在OSCC组织中的表达显著下调,且与OSCC的分级相关。miR-15b的前体能够使Cal27细胞的增殖能力明显下降,且能够诱导Cal27细胞发生凋亡。结论    miR-15b的异常表达与OSCC相关,miR-15b能够抑制OSCC细胞增殖并诱导其凋亡,在OSCC中发挥肿瘤抑制基因的作用。

Abstract:

Objective    To explore the expression of miR-15b in oral squamous cell carcinoma(OSCC) and the influence of miR-15b on OSCC Cal27 proliferation and apoptosis. Methods    Real-time PCR was used to detect the expression of miR-15b in OSCC and its adjacent tissues. The Cal27 cells were cultured and transfected with miR-15b precursor, then the proliferation and apoptosis ability of Cal27 cells were detected by MTT assay and double labeling flow cytometry method. Results    The miR-15b was down-regulated in OSCC tissues compared with the adjacent tissues and its expression showed a negative relationship with the grades of OSCC. The miR-15b precursor could significantly inhibit the Cal27 cells proliferation and induce the Cal27 cells apoptosis. Conclusion    The abnormal expression of miR-15b is involved in OSCC;miR-15b could inhibit the proliferation ability and induce apoptosis ability of Cal27 cells. The miR-15b acts as a tumor suppressor in OSCC.
